Rođen u Haarlemu, kao najstariji sin Franssa Pietersza de Grebbera (1573–1643), slikara i vezilca iz istog grada, De Grebber je odrastao u okruženju prožetom umjetnošću, budući i brat slikara Marije i Alberta. Svoje vještine stjecao je pod vodstvom vlastitog oca te priznatog Hendricka Goltziusa. Potjecao je iz katoličke obitelji umjetnika, a njegova je sestra Maria kasnije postala svekrva slavnog Gabriela Metsua. Njegov društveni krug bio je bogat kulturnim utjecajima; bio je prijatelj svećenika i muzikologa Jana Albertszoona Bana, čak je i pjesma za koju je skladio glazbu poznati haarlemski kompozitor Cornelis Padbrué bila povezana s njegovim životom. Iako je u 1632. godine službeno postao član Haarlemske eskadre Svetog Luke, on je kao umjetnik već deset godina bio aktivan i priznat. U tom dokumentu izlaže jedanaest najvažnijih pravila koja je smatrao ključnim za klasicističke slikare. Iako klasicisti nisu slijepo vjerovali tim pravilima, ona su se u praksi strogo poštavala, jer su većina njih preuzeta iz mannerističkog djela "Schilder-boeck" Karla van Mandera, u kojem se povijesna slika smatrala najvišom vrlinom u hijerarhiji žanrova.
